Tippy Tinkletrousers ( Professor Pippy Pee-Pee Poopypants; he changed his name after he went to jail, not by the chart) He bears similarities to Superman's archenemy, Lex Luthor. Both of them are intelligent scientists that, though without superpowers, are typically a threat to their foes.At the southwest of Greenland, a scientist from the fictional country of New Swissland (with a foreign culture where everyone has a silly name) named Professor Pippy P. Poopypants goes to the United States to demonstrate how his Shrinky-Pig 2000 and Goosy-Grow 4000 can help the world by reducing garbage and increasing food, but everyone laughs at Poopypants' silly name rather than taking him seriously, which is a constant annoyance to him. Meanwhile, Professor Poopypants finds a ad to teach at the school and applies for the job, thinking children to be kind and sweet-hearted, but they spend days laughing at his silly name instead of learning anything. The professor only gets them interested by building a robot that makes gerbils jog along with them, but the interest is short-lived when George gets him to reveal his middle name (pee-pee). Sometime later, Ms. Ribble reads The Pied Piper of Hamelin, which inspires George and Harold to make a comic about the Professor trying to take over the world using an army of gerbils in suits, which makes Professor Poopypants furious and destroys the last of his sanity. There is an apology and a giveaway at the end of the book, aimed at people who were "traumatized" by the new names inflicted on them by Professor Poopypants.

He got a job at Jerome Horwitz Elementary School in Piqua, Ohio as a science teacher, replacing Mr. Fyde who resigned. Everybody laughed at his name. He soon invented the Gerbil Jogger 2000 and showed it to the class. Everyone was impressed by it until they found out that his middle name was Pee-Pee, thanks to a snide remark from George and Harold. Later, he read a comic that the boys had wrote called "Captain Underpants and the Pied Pooper of Piqua". It destroyed his last bit of sanity and made him go crazy with revenge. In reference to their names, the Shrinky-Pig 2000 looks like a pig, and the Goosy-Grow 4000 resembles a goose. He then takes Mr. Krupp and breaks both of them out as he wishes to find George and Harold and get revenge on Captain Underpants for throwing him in prison.

On the advice of George and Harold, he changes his name so no one will make fun of it anymore (which he hadn't thought of until George noted that to him). Sadly, he chooses his maternal grandfather's name, Tippy Tinkletrousers, which the people at jail make fun of. George, Harold and their classmates tease the professor mercilessly about his name. He decides to impress them by bringing in an invention called the Gerbil Jogger 2000. It’s a small, robot-like device with an egg-shaped compartment at the top. When he inserts a gerbil, the long legs of the Gerbil Jogger 2000 spring into action. A student named Robbie Staenburg is mentioned as a student whose name was changed; Robbie Staenburg is the son of Dav Pilkey's agent. On the inside of the cover it shows Professor Poopypants crossing out Dav Pilkey's name and writing the silly equivalent (Gidget Hamsterbrains). Fourth-graders George Beard and Harold Hutchins have created their own comic superhero named Captain Underpants. They begin this book by drawing a comic for readers that recaps how they developed the character. They also show how they accidentally brought it to life when they hypnotized their principal. Now whenever someone snaps his or her fingers, Mr. Krupp turns into Captain Underpants and fights crime wearing only his briefs and a curtain for a cape. Near the end, Dr. Diaper makes a cameo appearance in the jail where the professor is sent to at the very end. He, along with many others in the jail, are seen laughing at Professor Poopypants's unbearably funny new name. According to his resume in the film, his birthday is on April 1, 1953, likely as a hint since April 1st is April Fools' Day, and it could be an indirect reference to him being a fool.Before Professor Poopypants is then hauled off to jail. George then questions Professor Poopypants wouldn't it have been smarter to try changing his own name instead of forcing the rest of the world to change theirs. The surprised professor notes he never thought of that. As per George's advice, Professor Poopypants changes his name so no one will make fun of it anymore. Sadly, he chooses his grandfather's name (on his mother's side), Tippy Tinkletrousers, which Dr. Diaper and the inmates at jail facility make fun of him (including the guards). Professor Poopypants is the main antagonist of the film. Professor Poopypants is first seen looking at a poster advertising a position for a science teacher at Jerome Horowitz Elementary School, and says it would be the perfect cover for his plan. He was the only character in the film to have his name changed from his book counterpart. In the movie, his full name is Professor Pee-Pee Diarrheastein Poopypants Esquire.
