About this deal

DNA extraction and sequencing of these modern and museum samples were conducted in the modern laboratory at the Archaeology Department of Durham University, following standard protocols (Supplementary Note 4). The cytb region was amplified in 10 overlapping fragments and a variety of primer combinations was used depending on the nature of the sample (Supplementary Table 6). The sequencing reaction was carried out by the DNA Sequencing Service at the School of Biological and Biomedical Sciences at Durham University. The sequencing chromatograms were edited manually, subsequently assembled, and a consensus sequence per individual exported using Geneious R6 version 6.0.6 ( https://www.geneious.com). Standard anti-contamination guidelines were followed. We successfully amplified cytb sequences from 202 of 292 samples. NaF was injected intravenously via a tail vein into male (n = 15, 32.3 ± 6.4 MBq) and female rats (n = 18, 32.2 ± 4.5 MBq) under brief isoflurane/oxygen anaesthesia. The rats (n = 6 for each time point) were sacrificed via cardiac puncture under increased anaesthesia at 15, 30, 60, 120, 240, or 360 min after injecting the tracer. Blood, urine, and organs of interest were removed, weighed, and measured for 18F-radioactivity in a NaI(Tl) well counter (3 × 3-inch, Bicron, Newbury, USA). The binding of [ 18F]fluoride to plasma proteins was determined by ultrafiltration, as previously described [ 11]. Rat populations are stable around much of the world. One of the largest threats to rats is habitat loss. Our finding that [ 18F]fluoride did not bind to plasma proteins was reported previously. This property is considered an advantage of [ 18F]fluoride, compared to [ 99mTc]diphosphonate, which showed 25–70% binding to plasma proteins [ 2]. Due to the lower molecular weight of fluoride compared to phosphonates, [ 18F]fluoride has a higher single-passage extraction efficiency compared to phosphonates [ 2]. The [ 18F]fluoride uptake we measured in bone marrow was consistent with the findings of Blake et al. [ 2], who speculated that the uptake in red blood cells might reflect the uptake of immature erythrocytes in bone marrow. That fascination with making the most of the leftover bits from a dig spurred Swift and her colleagues to analyze 145 rat bones from three Polynesian island systems in the Pacific. The rodents, considered invasive, disease-carrying pests in life, proved surprisingly useful in death. By measuring the chemical composition of the rat bones, the researchers could make inferences not only about what humans were eating around 2,000 years ago, but also how their early residence on the islands—Mangareva, Tikopia and Ua Huka (also known as Marquesas)—shaped the environment. The cytb region of the mitochondrial genome haplotypes were extracted using MEGA7, and combined with modern cytb haplotypes from previous publications 10, 24, 69, 70 and this study.
