About this deal

Determination of starch: Starch content was determined spectrophotometrically using the method by Thayumanavan and Sadasivam 29. In this method, about 250 mg of dry sample was homogenized in 80% ethanol to remove the sugar. The sample was then centrifuged at 5000 rpm for 5 min and then the residue was retained. After that, add 5 mL of cold distilled water and 6.5 mL of 52% perchloric acid were added to the residue. Then the solution was centrifuged and the supernatant separated and then filtered with filter paper No. 5 (Whatman). The processes were repeated until the supernatant was made up to 100 mL. A 100 μL of the supernatant was added to distilled water until the volume became 1 mL.
